The once great sketch comedy show 'Studio 60' is suddenly close to cancellation due not only to it's less than stellar on-air product. But another, more unexpected type of action, it's Executive Producer's on-air mental meltdown. So enter, NBS Chairman of the board, Jack Rudolph and a new network president, looking to put her mark on the struggling network, Jordan McDeere. This results in the hiring Danny Tripp and Matt Albie to replace the former Executive Producer and hopefully save the show.
The Studio 60 cast and crew continue to deal with the fallout of Wes' on-air breakdown, as well as adjusting to Matt and Danny's management. Harriet finds out that Matt is sleeping with Jeannie, a cast member and writer. Meanwhile, Jeannie stresses when a sketch she pitched doesn't go over as well as she hoped; and the team scrambles to find a musical guest when the lead singer for The White Stripes catches tonsillitis.
A network focus group sends a scare through the company that the ratings success of Matt (Matthew Perry) and Danny's (Bradley Whitford) first show might have been a one-time thing.
When it's discovered that the show has inadvertently plagiarized another comedian's material during a sketch, the team scrambles to break into the taped West Coast feed to correct their error.
While the cast goes through rehearsals with Lauren Graham and Sting, a reporter (guest star Christine Lahti) tries to get the story on Matt (Matthew Perry) and Harriet (Sarah Paulson) as Jordan (Amanda Peet) passes on a lucrative but tasteless new reality series.
A drunken Jordan (Amanda Peet) makes friends with the cast during an after-show party, Danny (Bradley Whitford) tries to get Matt's (Matthew Perry) mind off Harriet (Sarah Paulson) with the help of three sexy women, Simon (D.L. Hughley) makes a plea to hire more black writers and Cal (Timothy Busfield) deals with a mysterious man backstage (guest star Eli Wallach).
An unusual collection of people have to go to a small town in Nevada to get Tom (Nathan Corddry) out of jail in time for his show that night. Guest star John Goodman plays a judge who's not a fan of Studio 60.
Jack has to get Tom out of a small town jail to make it back for his show that night as tension heats up between Matt and Harriet.
After the show comes down, Matt discover that Ricky and Ron are planning to leave and take the writing staff with them. Harriet contemplates doing a lingerie spread in a magazine and Jordan and Danny wrestle with budget cuts.
The cast has to get through the show despite being ravaged by a virus -- and Matt has to get through the week with a new and much smaller writing staff. Jordan tells Danny that she's pregnant.
With a holiday show on the horizon, Matt is determined to bring the Christmas spirit to Studio 60. Which works romantic wonders for him. Meanwhile, Danny has to confront his true feelings for Jordan.
Danny continues his pursuit of Jordan while Matt competes on an online bid for a date with Harriet. Jack and Wilson must deal with their pending situation about Macau, while Jordan meets with Hallie Gallway regarding a new reality show pitch.
Danny and Jordan cannot get down from the roof of the theater; staff members attend a dinner honoring Harriet; Harriet and Matt's relationship begins to unravel.
